Purification and detoxification of trypsin-activated type E toxin. J. Bacteriol. 82:66-71. 1961.-A procedure was described for production, purification, and conversion to toxoid of the Clostridium botulinum type E trypsin-treated toxin. A medium containing 1.5% trypticase, 0.5% yeast extract, 0.075% cysteine hydrochloride, and 1.0% glucose consistently yielded activated culture toxicities of 300,000 mouse intraperitoneal ld(50)/ml. Purification of the toxin was accomplished by precipitation with ammonium sulfate, extraction with calcium chloride, and reprecipitation with ethanol in the cold. The resulting partially purified toxin had a specific activity of 45,000,000 ld(50)/mg nitrogen. Purified toxins were converted to toxoid by incubation with formalin and adsorbed on aluminum phosphate. Good immune responses were obtained to the toxoids in mice, guinea pigs, and rabbits.

Studies on immunity to toxins of Clostridium botulinum. III. Preparation, purification, and detoxification of type E toxin.
Although type E strains of Clostridium botulinum have been implicated in human botulism since 1936, no studies of the isolation or characterization of the toxin have been reported. Crude toxoids have been prepared by various workers for immunization of animals and preparation of antitoxin. Barron and Reed (1954) presented a systematic study of methods for preparation of crude alum precipitated ...
متن کاملExpression and Purification of Neurotoxin-Associated Protein HA-33/A from Clostridium botulinum and Evaluation of Its Antigenicity
Background: Botulinum neurotoxin (BoNT) complexes consist of neurotoxin and neurotoxin-associated proteins. Hemagglutinin-33 (HA-33) is a member of BoNT type A (BoNT/A) complex. Considering the protective role of HA-33 in preservation of BoNT/A in gastrointestinal harsh conditions and also its adjuvant role, recombinant production of this protein is favorable. Thus in this study, HA-33 was expr...
متن کاملThe antibody response of cattle to Clostridium botulinum types C and D toxoids.
The resistance of cattle with varying serum-antitoxin titres was determined by per os challenge. The results proved that a solid immunity can be produced against C. botulinum toxins C1 and D. The immune response of cattle to various quantities of C. botulinum C1 and D toxoids, aluminium-phosphate-adsorbed and in water-in-oil emulsion was investigated. The response to antigen in water-in-oil emu...
متن کاملImmunogenic and Protective Potentials of Recombinant Receptor Binding Domain and a C-Terminal Fragment of Clostridium botulinum Neurotoxin Type E
Clostridium Botulinum Type E neurotoxin heavy chain consists of two domains: the translocation domain asthe N-terminal half and the binding domain as the Cterminal half (Hc). One effective way to neutralize botulinum neurotoxin is to inhibit binding of this toxin to neuromuscular synapses with antibodies against binding domain. Two synthetic genes, coding for Hc (the full length binding d...
متن کاملEstablishment of a monoclonal antibody recognizing an antigenic site common to Clostridium botulinum type B, C1, D, and E toxins and tetanus toxin.
The partial amino acid sequence of the light-chain (Lc) component of Clostridium botulinum type C1 toxin was determined. The sequence was quite similar to those of the other types of botulinum and tetanus toxins. Nine monoclonal antibodies against botulinum type E toxin were established by immunizing BALB/c mice with type E toxoid or its Lc component. Six antibodies reacted with the heavy-chain...
